Standard addition graph. Allicin is a ~0.4wt%component in garlic with antimicrobial and possibly anticancer and antioxidant activity. It is unstable and therefore difficult to measure. An assay was developed in which the stable precursor alliin is added to freshly crushed garlic and converted to allicin by the enzyme alliinase found in garlic. Components of the garlic are extracted and measured by chromatography. The chromatogram shows standard additions reported as mg alliin added per gram of garlic. The chromatographic peak is allicin from the conversion of alliin.

(a)The standard addition procedure has a constant total volume. Measure the responses in the figure and prepare a graph to find how much alliin equivalent was in the unspiked garlic. The units of your answer will be mg alliin/g garlic. Find the 95%confidence interval, as well.

(b) Given that2molof alliin are converted to1molof allicin, find the allicin content of garlic (mg allicin/g garlic) including the95%confidence interval.

Short Answer

Expert verified

(a) The allilin equivalent in unspiked garlic was graphed, and the95% confidence interval was estimated as8.210.62mgalliin/g.

Step by step solution

01

Step 1:Given data of the question

Allicin is a ~0.4wt%

component in garlic with antimicrobial

The chromatographic peak is allicin from the conversion of alliin.

95%of confidence interval have to be found.

2molof alliin are converted to 1molof allicin

02

Equation to draw the necessary graph

It is necessary to graph the allilin equivalent in unspiked garlic and get the 95%

confidence interval.

Concept introduction:

The graph will be drawn using the below equation: role="math" localid="1654855448601" Plotls+x+XVV0Functiontoplotony-axisversuslx+lxXiSiVsV0functiontoplotonx-axis

HereX-intercept isXi

03

Quantity of alliin equivalent in the unspiked garlic

(a)

The allilin equivalent in unspiked garlic had to be graphed, and the 95%confidence interval was computed as 8.210.62mgalliin/g.

In the above spreadsheet x-axishas spike mg/gandls+xare plotted.

Figure 2 shows X-intercept=-bm=8.20006is the standard deviation of localid="1654856621530" X-intercept=8.210.62mgalliing/ggarlic.

The allilin equivalent in unspiked garlic was graphed, and the95%confidence interval was estimated as8.210.62mgalliing/ggarlic.

04

Step 4:The allicin content of garlic

(b)

Interpretation Introduction

Interpretation:

When 2molof alliin is converted to 1mol, the allicin content of garlic mgalliin/ggarlicmust be computed.

In the first figure, preadsheetx-axishas spike mg/gandls+xare plotted.

In the figure 3,the quantity of allicin in garlic ,when one mole of allicin is given by two moles of allin is:

12162.3/177.38.210.62mg=3.760.28mgalliicin/g

The amount of allicin in garlic was determined as 3.80.3mgallicin/g.

Olympic athletes are tested to see if they are using illegal performance-enhancing drugs. Suppose that urine samples are taken and analyzed and the rate of false positive results is 1 %. Suppose also that it is too expensive to refine the method to reduce the rate of false positive results. We do not want to accuse innocent people of using illegal drugs. What can you do to reduce the rate of false accusations even though the test always has a false positive rate of 1 %?

Chloroform is an internal standard in the determination of the pesticide DDT in a polarographic analysis in which each compound is reduced at an electrode surface. A mixture containing 0.500mMchloroform and0.800mMDDT gave signals of15.3渭础for chloroform and10.1渭础for DDT. An unknown solution(10.0mL)containing DDT was placed in a100-mLvolumetric flask and10.2渭尝of chloroform (FM 119.39, density=1.484g/mL)were added.
