Nucleosomes are crucial structures in the cell nucleus that facilitate the compact packaging of DNA. Think of them as tiny, bead-like units, where DNA strands are wrapped around a core of proteins called histones. Specifically, each nucleosome consists of a segment of DNA approximately 146 base pairs long wound around a core made of eight histone proteins, known as a histone octamer.
This configuration makes nucleosomes about 11 nanometers in diameter. Nucleosomes serve not just as packaging units but also play a critical role in regulating gene expression by controlling which parts of the DNA are accessible.

Histone proteins are essential to the organization and stability of chromatin. They act as spools around which DNA winds, allowing incredibly long DNA molecules to fit into the small confines of the cell nucleus. There are five main types of histone proteins: H1, H2A, H2B, H3, and H4. Together, they form the core around which DNA coils to create a nucleosome.
These proteins are relatively small, measuring approximately 5.5 nanometers, and are positively charged, which helps them bind effectively to the negatively charged phosphate backbone of DNA. This binding is crucial for the stable formation of nucleosomes, enabling efficient DNA packing. Additionally, histones play a vital role in gene regulation because chemical modifications to histones can control DNA accessibility.

Chromatin is the fundamental structure of DNA within the cell nucleus, comprising DNA molecules tightly coiled around histone proteins to form nucleosomes. These nucleosomes further coil and fold to form chromatin fibers, which can be loosely organized in a non-condensed form known as euchromatin or tightly packed into heterochromatin.
This structure is dynamic, allowing chromatin to alter its arrangement during various cellular processes, such as replication and transcription. During interphase, chromatin fibers measure about 30 nanometers in diameter, but this can change during cell division. Chromatin not only serves as a physical structure but also plays a key role in regulating gene expression and safeguarding DNA integrity.
