Adenylate cyclase plays a pivotal role in the signal transduction pathways within cells. It is an enzyme responsible for converting ATP into cyclic AMP (cAMP), a secondary messenger that is essential in many biological processes. In the context of the lac operon, adenylate cyclase activity is essential for the production of cAMP. This molecule activates the catabolite activator protein (CAP), which enhances the transcription of the lac operon genes when glucose levels are low. Without active adenylate cyclase, cAMP levels remain low, preventing CAP from binding to the promoter and stimulating transcription. This explains why mutations that impair adenylate cyclase result in reduced transcription of the lac operon, regardless of lactose availability.

Cyclic AMP (cAMP) is a vital secondary messenger in many cellular processes, mediating responses to hormonal signals and environmental changes. In bacteria, cAMP is involved in regulating the lac operon. Its levels in the cell are inversely related to glucose concentration. When glucose is scarce, cAMP levels rise, enabling cAMP to bind to CAP. This coordination ensures the bacterium prioritizes glucose utilization over lactose metabolism, optimizing energy efficiency.
