Prions are unique biological entities that challenge traditional notions of what it means to be infectious. To fully appreciate how prions propagate, we must first understand their structure. Prions are primarily composed of a protein known as PrP^C (cellular prion protein) that ordinarily exists harmlessly in animal tissues. However, prions represent a malformed version of this protein, aptly designated PrP^Sc (scrapie protein), associated with disease.

Unlike pathogens with nucleic acids, prions lack DNA or RNA; they are simply protein molecules with a particular three-dimensional shape. This misfolded conformation is highly stable and resistant to degradation, a feature that partly explains why prions are formidable agents of disease. Their structure allows them to induce conformational changes in the normal, properly folded PrP^C, catalyzing a domino effect which alters the structure of neighboring proteins and propagating the infectious state.

When discussing infectious agents, we typically refer to organisms with genetic material, such as bacteria, viruses, or fungi, capable of replicating by producing copies of themselves. Prions defy this conventional categorization because they are neither cellular organisms nor do they carry genetic material. Nonetheless, they are infectious because of their remarkable ability to recruit and convert the body's own proteins.

The infectious nature of prions is associated with several fatal neurodegenerative diseases, such as Creutzfeldt-Jakob disease in humans and bovine spongiform encephalopathy (more commonly known as 'mad cow disease') in cattle. These diseases are characterized by the accumulation of prion proteins in the brain, leading to cell death and tissue damage, which ultimately results in the deterioration of brain function.

At the heart of prion replication lies the phenomenon of protein misfolding. Proteins are the workhorses of our cells, and their function is inextricably linked to their shape. Normally, proteins fold into precise structures to perform their biological roles effectively. Prion diseases arise when the normal prion protein (PrP^C) undergoes a conformational change to become misfolded (PrP^Sc), thereby becoming pathogenic.

The misfolded prion then acts as a template that promotes the transformation of other normal proteins into the abnormal configuration. This autocatalytic cycle leads to an accumulation of misfolded proteins, forming aggregates that the cell cannot easily break down. These aggregates disrupt cellular function and lead to the symptoms of prion diseases.
