Prion proteins are uniquely capable of inducing their abnormal form onto normal variants of the same protein. This process is referred to as prion protein misfolding. Normally, prion proteins exist harmlessly in the body, particularly in the brain. However, when these proteins misfold, they adopt a structure that is not only abnormal but also infectious.

Imagine the prion protein like a complex 3D puzzle, and when it's correctly assembled, it fits perfectly into the body's daily operations. Misfolding would be akin to forcing a piece into the wrong place, causing a cascade of errors. This misfolded structure is remarkably stable and can convert other normal prion proteins into the misfolded, pathogenic form. The process is akin to a domino effect, where one misfolded protein triggers the misfolding of others, leading to a build-up of prions that the body cannot clear.

This accumulation of abnormal proteins disrupts normal cellular processes and eventually leads to the damage and death of neurons, manifesting as a variety of neurodegenerative symptoms in affected individuals.

Neurodegenerative diseases are a group of disorders characterized by the progressive degeneration of the structure and function of the nervous system. They are incurable and debilitating conditions that result in progressive degeneration and death of nerve cells, causing problems with movement or mental functioning. Examples include Alzheimer's disease, Parkinson's disease, and prion diseases such as Creutzfeldt-Jakob Disease.

These diseases have a wide range of symptoms but commonly include memory loss, impaired judgment, and motor coordination difficulties. Over time, individuals may lose the ability to move or speak. The progression of neurodegenerative diseases is relentless, and while some treatments can provide relief from physical or mental symptoms, there is no cure. Research suggests that misfolded proteins play a significant role in these diseases—prion diseases are the most direct example of this, where misfolded prion proteins cause direct damage to brain tissue.

Pathogenic prions are the infectious form of prion proteins that have misfolded and are responsible for a range of fatal neurodegenerative diseases, collectively known as prion diseases. This rogue form of the prion protein can trigger normal proteins to misfold, propagate the misfolded form, and result in a chain reaction.

Unlike most infectious agents like bacteria and viruses, pathogenic prions don't contain nucleic acids, meaning they lack DNA or RNA. Instead, they propagate by transmitting their abnormal protein folding to normal versions of the protein, a novel method of infection. The diseases caused by pathogenic prions can affect both animals and humans. In animals, bovine spongiform encephalopathy (BSE), commonly known as mad cow disease, is the most recognized prion disease. In humans, variant Creutzfeldt-Jakob Disease (vCJD) is linked to exposure to BSE. Their ability to spread and the absence of a standard immune response make pathogenic prions a particularly challenging subject for research and a concern for public health.
