Excision repair is a crucial mechanism that cells use to fix damaged DNA. This process targets abnormal or altered bases that may have occurred due to various damaging factors like ultraviolet light, chemicals, or radiation. Initially, specific proteins in the cell detect the damage in the DNA strand.
Once discovered, the cell employs different enzymes to remove the damaged section. This involves cutting out not only the altered base but also a stretch of the surrounding DNA. After this segment is excised, DNA polymerase, an enzyme, steps in to synthesize the correct sequence using the undamaged strand as a guide. The final step is the reattachment of this newly synthesized piece back into the DNA strand, a process known as ligation.
Excision repair is vital for preventing mutations that could interfere with cell function or lead to diseases.

Mismatch repair plays a critical role in correcting mistakes introduced during DNA replication. When the DNA is being replicated, errors can occur, like mispairing of the nucleotides (the building blocks of DNA) or small insertions and deletions that disrupt the base pairing.
Mismatch repair proteins are responsible for identifying these replication errors. Once the mismatch is detected, the incorrect piece of DNA is removed from the newly synthesized strand.
Subsequently, DNA polymerase inserts the correct nucleotides using the original (parental) DNA strand as a template. Finally, the DNA strand is sealed with the help of another enzyme, ligase, ensuring that the sequence is consistent and does not harbor errors.
Without mismatch repair, cells would accumulate mutations rapidly, leading to potentially harmful consequences like cancer.

During the process of DNA replication, the DNA double helix is unwound, and new strands are synthesized based on the original strands. However, this complex procedure is not foolproof. Sometimes, errors can occur, such as wrong base pairing or accidental insertion/deletion of bases.
Such DNA replication errors are natural and happen regularly, but if left unchecked, they can lead to genetic variations or serious mutations.
